Abstract

Antioxidants are compounds that can prevent or slow down oxidation reactions caused by free radicals, particularly in lipid oxidation, by donating electrons to neutralize them. This study aimed to evaluate the antioxidant activity of ethyl acetate and n-hexane fractions of pomegranate (Punica granatum L.) leaves compared with vitamin C as a standard, and to determine their IC₅₀ values against DPPH free radicals. Extraction was carried out by maceration using 96% ethanol, followed by fractionation with ethyl acetate and n-hexane. Antioxidant activity was quantitatively tested using the DPPH (2,2-diphenyl-1-picrylhydrazyl) method, and absorbance was measured with a UV-Vis spectrophotometer. The results showed that the IC₅₀ value of the ethyl acetate fraction was 15.646 ppm, the n-hexane fraction was 73.080 ppm, and vitamin C was 11.87 ppm. Based on these findings, the ethyl acetate fraction demonstrated very strong antioxidant activity, while the n-hexane fraction showed strong activity.
